ATAC-sequencing of genetically engineered intestinal organoid lines modeling colorectal cancer (CRC) progression

To characterize changes in chromatin accessibility acorss the CRC malignancy continum, wild type organoids (WT), ApcKO (A) and ApcKO::KrasG12D::Smad4KO::Trp53KO (AKSP) tumoroids were subjected to bulk ATAC-seq. We then performed differential accessibility analysis using data obtained from ATAC-seq of the indicated organoid lines. Overall design: Wild type intestinal organoids (WT), ApcKO (A) and ApcKO::KrasG12D::Smad4KO::Trp53KO (AKSP) tumoroids were collected for nuclei isolation and DNA transpostion when they reached confluence.
