Mitochondrial DNA sequence of cell line mixtures MCF-7 and MDA-MB-231

Evaluation of limit of detection low-frequent mitochondrial DNA variants by Pacific Biosciences' Single-Molecule Real-Time sequencing. Sample mixtures containing cell lines MCF-7 and MDA-MB-231 were prepared. To determine detection limits, we prepared mixtures of the cell lines – considering MDA-MB-231 as the mutant variant – to generate samples with allele frequencies of 0%, 0.001%, 0.01%, 0.1%, 1% and 10% variant. Also, a sample containing 100% MDA-MB-231 was sequenced by Single-Molecule Real-Time sequencing as well as Illumina sequencing-by-synthesis.
