Genomic and transcriptomic study of aneuploid Candida albicans strains.

In this study we karyotyped 38 aneuploid Candida albicans strains, along with a diploid, tetraploid, and three clinical isolates using double digest restriction-site associated DNA sequencing (ddRAD-seq), and preformed transcriptional profiling using RNA-sequencing. Strains were grown in yeast peptone dextrose (YPD) liquid medium to an OD of 1.0 at 30 degrees Celsius, at which point cultures were split into two cryovials, snap frozen in liquid nitrogen, and stored at -80 degrees Celsius. DNA and RNA were extracted directly from the frozen pellet for ddRAD-seq and RNA-seq library construction. The goal of this project was to identify any common transcriptional responses to aneuploidy and to study expression patterns on imbalanced chromosomes.
