DNA Repair Gene Expression Associated with Oxidative Stress Responses

The transcriptomic effect in DNA repair genes after long-term oxidative stimuli was assessed as follows: C2C12 cells (ATCC, CRL-1772) were exposed to a chronic pro-oxidant treatment outline performed with daily 1-hour addition of glucose oxidase (GlucOx, Sigma G6641) to produce hydrogen peroxide at a concentration of ~50 μM. Gene expression levels were obtained from C2C12 cells by qRT-PCR-based microarray (QIAGEN PAMM-042Z) performed in both control cells (NT) and cells exposed to chronic GlucOx treatments (ADP). Cycle threshold (Ct) values obtained for three independent experiments are shown in Table 1.
