This project utilises ChIP-seq to analyse the seasonal dynamics of H3K27me3 and H3K4me3 in the natural environment. Field sampling was conducted monthly for a year in a natural population of perennial Arabidopsis halleri subsp. gemmifera in Hyogo prefecture, Japan. The leaf samples were cross-linked by formaldehyde in the field and transferred to the laboratory. To analyse the seasonal dynamics of the two histone modifications, ChIP was conducted for the samples collected at 12 time points from 6 November 2012 to 24 September 2013, using anti-H3K27me3 and anti-H3K4me3 antibodies. ChIP DNA for each time point was sequenced by HiSeq2500 including two biological replicates.
