We establish Enhancer-Promoter loop of Dppa5a in pre-iPSC by a chemical-inducible system to promote the establishment of pluripotency. Overall design: The SadCas9-ABI and SpdCas9-PYL , both fused with IDR-truncated mutant of JMJD2C (Mut), Mut-hIDR1 or hIDR1, were targeted to enhancer and promoter of Dppa5a. We induced fusion of JMJD2 phase at enhancer and promoter of Dppa5a by adding ABA for 96h and washed-out. The E-P loop of Dppa5a was evaluated at 168h by 4C-seq with bait in the promoter region of Dppa5a.
