Scope of the article:This protocol describes the general processes and quality control checks necessary for preparing healthy adult single cells in preparation for droplet-based, high-throughput single cell RNA-Seq analysis using the 10X Genomics' Chromium System. We also describe sequencing parameters, alignment and downstream single-cell bioinformatic analysis.
