PARP1 is a member of various DNA repair pathways - main target for anti-cancer treatment. We evaluated the effect of topotecan and TDP1 Inhibitor in HEK293 wild-type and PARP1 knockout cells on the gene expression Overall design: Knockout HEK293A cell line was generated using the CRISPR/Cas9 method. gRNA were design against region with constitutive protein-encoding exons of Parp1 gene. Then, we performed comparative gene expression profiling analysis of RNA-seq data for HEK293 wild type cells and Parp1 KO under influnce of topotecan, OL9-119 and its combination.
