Mechanosensitive calcium channels and integrins coordinate the reprogramming of colorectal cancer cells into a fetal-like state

Plate-based scRNA-seq data of murine CRC organoids that were embedded in either Matrigel-or Collagen I matrices and harvested after 1 day. Overall design: Murine colorectal cancer organoids (genetic background VillinCre-ERT2; ApcFL/FL; KrasLSL-G12D/+; Tp53KO/KO; R26R-Confetti; Lgr5DTR-eGFP) were cultured for 3 days in Matrigel. The CRC organoids were isolated from the Matrigel drops using dispase treatment and re-embedded in either Matrigel or Collagen I matrices. After 1 day the organoids were subjected to fluorescence associated cell sorting and analyzed by plate-based single-cell RNA sequencing.
