Saccharomyces cerevisiae raw sequence reads for Bar-Seq and QTL analysis of pathogenicity

The baker’s yeast, Saccharomyces cerevisiae, is widely used for research and industry but also is known as an opportunistic pathogen of humans. Here we leveraged a number of yeast resources and a model infection system, inoculation into the greater waxmoth, Galleria mellonella, to provide a genome-wide analysis of pathogenicity factors. We performed three experiments involving sequences in this submission: 1) serial passaging of wild-type strains followed by quantification of relative pathogenicity by sequencing strain specific barcodes (Bar-seq), 2) serial passaging of the deletion collection followed by quantification of relative pathogenicity by Bar-seq, 3) Bulk segregant analysis of cross between virulent and less virulent strains. We identified genes involved in cell wall integrity, mitochondrial function, and tyrosine metabolism as being critical for pathogenicity in our model host.
