We reasoned that when livers grow beyond their normal size, some growth regulating secreted factors will be suppressed and others will be overexpressed. To obtain overgrown livers, we used genetic models involving YAP or MYC overexpression We the identified differentially expressed genes in both models and filtered out secreted factors by bioinformatically defined secreted protein list. Col-TRE-YAPS127A; Rosa26-LSL-rtTA transgenic mice were given AAV-TBG-Cre between 8-10 weeks of age to induce rtTA expression in the liver. Upon dox administration, YAPS127A was induced for two or four weeks, then livers were collected. We also used the LAP-tTA; TRE-MYC model of inducible liver specific MYC overexpression. MYC was induced at ~E8 by withdrawing dox water from pregnant mothers; livers were collected from offspring 4-weeks after birth.
