本研究设计并构建了动力学可精确调控的可编程DNA分子开关,通过将可重构茎环结构作为开关元件插入立足点区域与分支迁移区域之间,基于分子内运动的构像转换可实现4个数量级内的链置换反应动力学调节,为功能性DNA分子电路的设计构建提供了灵活可控的基础构筑模块。通过更换开关区域组件进一步赋予核酸开关分子以环境响应性行为,实现了响应光化学调控的核酸分子逻辑电路的设计构建。以质子响应性三链体基元结构为开关区域,将其插入toehold区域与识别区域之间,构建了基于三链体的核酸开关分子,可实现基于pH响应的DNA链置换反应。此外,结合光致变色分子螺吡喃,本研究工作进一步实现了可响应光刺激以变换构像的核酸开关分子。以光化学调控的核酸开关分子为基元结构,结合多种分子结构设计,实现了可完成多种逻辑计算功能的核酸分子电路,并成功赋予电路以光化学响应性行为。本数据集是关于光/pH响应型的DNA逻辑电路探针的合成和性能表征数据,主要包括非变性聚丙烯酰胺凝胶电泳数据、荧光光谱数据、理论计算数据。
In this study, programmable DNA molecular switches with precisely tunable kinetics were designed and constructed. By inserting reconfigurable stem-loop structures as switch elements between the toehold domain and the branch migration domain, conformational conversion based on intramolecular motion enables kinetic regulation of strand displacement reactions across four orders of magnitude, providing flexible and controllable basic building blocks for the design and construction of functional DNA molecular circuits.
By replacing the components of the switch region, the nucleic acid switch molecules were further endowed with environmental responsiveness, enabling the design and construction of nucleic acid molecular logic circuits regulated by photochemical stimuli. Using a proton-responsive triplex motif as the switch region and inserting it between the toehold and recognition domains, a triplex-based nucleic acid switch was constructed, which can realize pH-responsive DNA strand displacement reactions. In addition, combined with the photochromic molecule spiropyran, this study further developed nucleic acid switches that can alter their conformation in response to light stimulation.
Using photochemically regulated nucleic acid switches as basic motifs and integrating various molecular structure designs, this study realized nucleic acid molecular circuits capable of executing multiple logical computation functions, and successfully endowed these circuits with photochemical responsiveness.
This dataset comprises synthesis and performance characterization data for light/pH-responsive DNA logic circuit probes, primarily including non-denaturing polyacrylamide gel electrophoresis (PAGE) data, fluorescence spectroscopy data, and theoretical calculation data.
