A comparative metabarcoding study of mock soil nematode communities

Two nematode sets, composed of DESS treated and DESS-untreated (fresh) individuals, were prepared. These sets differed slightly in species composition and number of individuals. 6 replicates of each set were prepared. The first set was composed only of nematodes originally preserved in DESS. The second set of the composed nematode taxa consisted of nematodes picked out of water suspensions, immediately after they were extracted from soil (roots or leaves). Three replicates of each nematode set were used for DNA isolation using one of the two tested isolation kits. As a result, 12 samples were prepared (representing 2 sets of soil nematode taxa, isolated with 2 methods in 3 repeats each). From each of the 12 DNA isolates 18S and 28S rDNA reagions were amplified and subsequetly sequenced using the MiSeq sequencer (Illumina).
