Understanding the high L-threonine production in Escherichia coli TWF001 using two phases transcriptomics

The transcriptional level of 1739 genes and 2362 genes were changed significantly in logarithmic phase and stationary phase respectively by the transcriptome comparison of TWF001 and W3110. The L-methionine synthesis ability was weakened according to the down-regulation of metABC and the same as L-glycine synthesis ability through down-regulation of ltaE, tdh, kbl and L-lysine synthesis ability through up-regulation of argD. The up-regulation of transporter genes rhtA promoted the L-threonine secretion capability of TWF001. The up-regulation of the genes coding for ribosomal proteins were beneficial for L-threonine synthesis in E. coli. Some genes related to the L-threonine producing were achieved, argP, hfAB, gcvPT, gcvH, etc to guide our future work in TWF001. All information achieved was great significance for the genome breeding of better industrial L-threonine producing strains.
