Regulatory T cell-derived TGF-β signaling governs the heterogeneity and abundance of tumor-infiltrating bystander CD8+ T cells

ATAC-seq library was prepared using Hyperactive ATAC-seq Library Prep Kit for Illumina (Vazyme, TD711) to study bystander P14 CD8+ T cells in tumor. Here, we further demonstrate that TME-infiltrating TBYS cells subpopulations, including TCM-like, TEM-like, TRM-like ones, exhibit distinct epigenetic programs. This study gives a epigenetic profiles of bystander P14 CD8+ T cells in tumor. Congenic CD45.1+ P14 CD8+ T cells were adoptively transferred into naive C57BL/6 recipients (CD45.2+), which were then infected with LCMV Armstrong and engrafted with MC38 cells on Day 60 post infection. On day 15 post tumor engraftment, P14 CD8+ T cells in tumor were sorted by FACS and analyzed using ATAC-seq.
