Cyclin D1 is one of the most important oncoproteins that drive cancer cell proliferation and associates with tamoxifen resistance in breast cancer. In order to determine whether LncRNAs bind to Cyclin D1 and regulate its function, MCF-7 cells with exogenous HA-tagged or untagged Cyclin D1 were established and subjected to RNA immunoprecipitation (RIP) using anti-HA antibody. RIP-Sequencing (RIP-seq) was then performed to identify the lncRNAs that specifically binds to HA-tagged Cyclin D1, we identify a lncRNA, DILA1, interacts with Cyclin D1 and is overexpressed in tamoxifen resistant breast cancer cells.
