Altered lipid metabolism contributes to lung inflammation and drives lung tumorigenesis. A key factor maintaining homeostasis is Sphingosine Kinase 1 (SPHK1), which phosphorylates sphingosine to the bioactive lipid sphingosine-1-phosphate. Interruption of this synthesis pathway in lung tumor cells promotes a pro-tumorigenic microenvironment and hence drives tumor progression. To better understand this process, we performed RNAseq analysis of SPHK1 proficient and deficient mouse lung tumor cells harboring KrasG12D mutations. Overall design: We established KrasG12D driven, p53 deficient lung tumor cell lines. Subsequently, we knocked out SPHK! In these cells. For RNAseq analysis, we harvested RNA before and after treatment witth 10% fetal calf serum.
