The goal is to identify genes required to activate WNT/beta-catenin signaling in response to WNT3A. RKO (human) cells expressing a fluorescence-based transcriptional reporter of WNT signaling strength was transduced with the genome-wide Brunello CRISPR library, treated with WNT3A conditioned media, and sorted for the bottom 10% population, which includes disruptions to positive regulators of the WNT signal transduction pathway.
