LBD CWOW single nucleus RNA sequencing pilot studies

In order to optimize nuclei preparation protocols and select an appropriate sequencing platform to perform single-nucleus RNA sequencing on a large cohort of archived postmortem brain tissue from dementia cases, 3 small pilot studies were conducted: 1) two nuclei samples were prepared/sequenced from the same case using two different nuclei enrichment steps (miltenyi anti-nucleus microbeads or miltenyi debris removal) and the PIPseq platform; 2) several samples from different nuclei preparations from the same case, using the PIPseq platform; and 3) a 2 case vs 2 control comparison between sequencing platforms 10X and PIPseq.
