资源简介:
Biofilm formation by bacterial strains can be visualised after growth in a 96-well plate (see images in related submission) where the addition of the dye, crystal violet, stains the biofilm formed a deep purple colour. The degree of biofilm formation can be measured spectrophotometrically, by resuspending the dye-stained biofilms in 30% (v/v) acetic acid. These measurements can be compared to that of the media control and plotted in order to visualise the extent of biofilm formation when strains are cultivated under the same conditions.Biofilm_CV_Plate 1: Excel sheet containing different tabs depicting the general information generated by the microtiter plate reader, the layout of the 96-well plate used (strains assigned to specific wells), and the photometric data. ‘CV’ denotes the use of the dye, crystal violet, in the visualisation of a biofilm formation. For Plate 1, cultures were grown in tryptic soy broth (TSB) for testing biofilm formation.Biofilm_CV_Plate 1_TSB+: Excel sheet containing different tabs depicting the general information generated by the microtiter plate reader, the layout of the 96-well plate used (strains assigned to specific wells), and the photometric data. ‘CV’ denotes the use of the dye, crystal violet, in the visualisation of a biofilm formation. For Plate 1_TSB+, cultures were grown in tryptic soy broth (TSB) prepared in a Red Sea salt solution (simulating seawater; 38.2 g/L) for testing biofilm formation.Biofilm_CV_Plate 2: Excel sheet containing different tabs depicting the general information generated by the microtiter plate reader, the layout of the 96-well plate used (strains assigned to specific wells), and the photometric data. ‘CV’ denotes the use of the dye, crystal violet, in the visualisation of a biofilm formation. For Plate 2, cultures were grown in TSB for testing biofilm formation.Biofilm_CV_Plate 2_TSB+: Excel sheet containing different tabs depicting the general information generated by the microtiter plate reader, the layout of the 96-well plate used (strains assigned to specific wells), and the photometric data. ‘CV’ denotes the use of the dye, crystal violet, in the visualisation of a biofilm formation. For Plate 2_TSB+, cultures were grown in TSB prepared in a Red Sea salt solution (simulating seawater; 38.2 g/L) for testing biofilm formation.
