We obseverd Wnt pathway activation in BrafV637E-induced mouse intestinal tumours. To analyse the mechanisms of Wnt pathway activation, we initially performed whole-exome sequencing of 20 Braf-mutant tumors. We have identified a number of mutations in known Wnt pathway genes. Shown are sequences spanning mutations that were subsequently confirmed by PCR-based amplification and 454 resequencing. Furthermore, we have resequenced the most frequently mutated Wnt pathway genes (Apc, Ctnnb1 and Lrp1b) in another set of 46 tumours. The sequence reads are presented.
