mRNA ac4C Profiling in NAT10-Regulated Colonic Epithelial Senescence

This study aims to characterize transcriptome-wide changes in N4-acetylcytidine (ac4C) RNA modification during colonic epithelial cell senescence. Using ac4C RNA immunoprecipitation followed by sequencing (acRIP-seq), we profiled ac4C-modified mRNAs in human colonic epithelial NCM460 cells under three conditions: (1) proliferating control cells, (2) Hydrogen-Peroxide-induced senescent cells (H-Sen), and (3) Hydrogen-Peroxide-induced senescent cells with NAT10 knockdown (H-Sen-Si). NAT10 is a known ac4C "writer" enzyme. Our data reveal the dynamic regulation of ac4C modifications during senescence and identify NAT10 as a key regulator of ac4C deposition on target mRNAs. This dataset provides new insights into the epitranscriptomic landscape of aging in intestinal epithelial cells.
