Melanomas are considered highly heterogenous in transcriptional state. We used single cell RNA sequencing (scRNA-seq) to analyze the diversity of melanoma cells as compared to primary melanocytes and nevus melanocytes. Sample 1 contain KIT postitive melanocytes sequenced directly ex vivo from human nevi. Sample 2 contains different human melanoma cell lines and primary human melanocytes grown in vitro. Overall design: Melanoma cell lines were grown in RPMI media supplemented with 10% FBS. NHMs were grown in 254 media supplemented with HGMS. Nevus cells were obtained by excising nevi and sorting for KIT positive cells using the protocol in PMID: 34475532
