Impact of dieldrin on transcription in Jurkat T cells

Dieldrin is a pesticide with a suspected impact on chromatin. Here, we have exposed Jurkat cells to dieldrin for 30 min. and examined by next generation sequencing the impact on transcription. This impact was compared to that of DMSO (solvent of the dieldrin) and that of PMA. Overall design: Jurkat cells were exposed to either DMSO, 100microM dieldrin, or 100microM PMA for 30 min. Total RNAs were isolated with a phenol-chloroform-based protocole. Stranded libraries were prepared by random priming, enrichched in poly(A), and then sequenced with a paired end protocole. A second series of libraries were also prepared with RNAs from DMSO and dieldrin-treated cells using a rRNA-depletion approach rather than a poly(A) enrichment. These libraries were sequences alike.
