To date, one primer set has been designed for phoD alkaline phosphatase in soil. However, these primers have been reported to have an amplification bias towards Alphaproteobacteria. This leads to a lower coverage of the large phoD diversity. This study consists of the comparison of the published primers with newly-designed primers targeting phoD alkaline phosphatase using 454-sequencing on six grassland soils.
