In order to find the downstream mechanism of peptide ADSCP2, we performed RNA-sequencing (RNA-seq) to examine the transcriptome change induced by ADSCP2. Differentially expressed genes were obtained by RNA-seq. Compared with the scramble control, 160 genes were up-regulated (fold change = 2, P < 0.05) and 170 genes were down-regulated (fold change = 0.5, P < 0.05) in the ADSCP2 group. Overall design: Primary hypertrophic scar fibroblasts were treated with 25 µM Sc or ADSCP2 in triplicates for 24 h and collected for transcriptome library preparation and sequencing (Illumina NovaSeq6000, in PE150 mode) by ShanghaiBio Corporation. Differentially expressed genes (DEGs) were found using edgeR and corrected with false discovery rate (FDR). Genes with a fold change of =2 and q value = 0.05 between two groups were identified as differentially expressed genes.
