Methylenetetrahydrofolate dehydrogenase 2 (MTHFD2), a central mitochondrial enzyme in one-carbon metabolism, is frequently overexpressed in multiple human cancers and has been implicated in tumor progression. However, its functional role and transcriptional regulation in cutaneous squamous cell carcinoma (cSCC) remain poorly understood. To address this, we performed stable knockdown of MTHFD2 in the SCL-1 cSCC cell line using shRNA, with non-targeting shRNA as a control. RNA sequencing was subsequently conducted to compare transcriptomic profiles between MTHFD2-deficient and control cells, aiming to identify differentially expressed genes and elucidate the molecular networks regulated by MTHFD2 in cSCC. This dataset provides a resource for further exploration of MTHFD2-driven mechanisms in skin carcinogenesis.
