To determine the changes in liver immune cells upon hepatic Bicd2 deficiency, we performed scRNA-seq to analyze the hepatic immune microenvironment in Alb-WT Bicd2fl/fl and Alb-Cre Bicd2fl/fl mice subjected to S-100 induced AIH model. Overall design: Alb-WT Bicd2fl/fl and Alb-Cre Bicd2fl/fl mice subjected to S-100 induced AIH model. To obtain hepatic immune cells, liver tissues were minced and grinded in pre-cold FACS buffer (PBS containing 1% FBS), followed by filtration through a 70 µm cell strainer. Lymphocytes were isolated through 40/70% Percoll (Cytiva, Cat# 17089109) by gradient centrifuging 800 × g for 20 min. Cells located at the inter-layer were collected. After removing red blood cells resuspended with ACK lysis buffer, living cells were sorted and pooled using the 10× Genomics platform and sequenced on an Illumina NextSeq sequencer.
