To determine the changes in liver immune cells upon hepatic Bicd2 deficiency, we performed scRNA-seq to analyze the hepatic immune microenvironment in Alb-WT Bicd2fl/fl and Alb-Cre Bicd2fl/fl mice subjected to S-100 induced AIH model. Overall design: Alb-WT Bicd2fl/fl and Alb-Cre Bicd2fl/fl mice subjected to S-100 induced AIH model. To obtain hepatic immune cells, liver tissues were minced and grinded in pre-cold FACS buffer (PBS containing 1% FBS), followed by filtration through a 70 µm cell strainer. Lymphocytes were isolated through 40/70% Percoll (Cytiva, Cat# 17089109) by gradient centrifuging 800 à g for 20 min. Cells located at the inter-layer were collected. After removing red blood cells resuspended with ACK lysis buffer, living cells were sorted and pooled using the 10à Genomics platform and sequenced on an Illumina NextSeq sequencer.
