To reveal transcriptome dynamics during adventitious root formation in a coniferous tree, C. japonica, we conducted custom microarry experiments. Three parts from cuttings of easy-to-root clone of C. japonica were collected at eight time points during adventitious root formation. The results revealed major turning points on transcriptome toward adventitious root formation and the expression behavior of genes related to carbohydrate, plant hormone and others suggested the important biological changes for adventitious root formation. Base of cuttings (underground), middle of cuttings (above ground) and shoots including needles were collected at 8 time points (0 hpe (hour post excision), 0 hpi (hour post insertion, about 2 hpe), 3 hpi, 1 dpi (day post insertion), 3 dpi, 1 wpi (week post insertion), 3 wpi and 6 wpi). At each time point, samples were collected from three cuttings as biological replicates.
