Liph, a gut-enriched Lipase H encoding gene, shows decreased expression during gut aging in both fruit fly and mouse. However, whether such evolutionary conserved Liph plays a protective role in gut aging remains unknown. In this study, RNA-seq was performed on gut tissue from fly model to identify changes in gene expression produced in whole guts after knocking down CG6295, the Drosophila ortholog of the mammalian Liph. In addition, we performed RNA-seq analysis on rat intestinal epithelial IEC-6 cells with Liph knockdown to investigate the role of Liph in mammalian intestinal aging. Overall design: To perform RNA-sequencing of the whole gut, flies from CG6295 RNAi-1 and control flies at 5 days after eclosion were prepared. The whole guts (30 per sample) were carefully dissected from each genotype by removing trachea and Malpighian tubules. We performed RNA-seq analysis on rat IEC-6 cells with Liph knockdown by two short hairpin RNAs (Liph_sh1, Liph_sh2). Three biological replicates for each sample.
