We established a micro-patterned respiratory epithelial cell culture system in vitro. In this culture system, various types of lung epithelial cells were identified. Human iPSC-derived lung progenitor cells were seeded onto micro-patterned plates and cultured in "DCIK+3i" (CHIR99021, SB431542 and Y27632) medium or "PAL" medium for 7 or 14 days to induce alveolar or airway epithelial cell colonies, respectively. These cells as well as their parental progenitor cells, were dissociated and subjected to single-cell RNA sequencing (scRNA-Seq) analysis.
