The identification of T cell targets to improve immunotherapies would be useful, but large scale CRISPR screens directly in T cells in vivo has been limited. Here, we developed a hybrid genetic screening system with adeno-associated virus (AAV) and the Sleeping Beauty (SB) transposon. We performed in vivo AAV-SB-CRISPR screens in primary murine CD8+ T cells to identify membrane protein targets that modulate anti-tumor activities in a mouse model of glioblastoma.
