Multi-parameter flow cytometry has advanced the phenotyping of immune cells allowing for assessment of multiple antigens at a protein level. However, the vast majority of optimized panels favors the analysis of T cells, and there are currently no panels which primarily assess the phenotype of murine myeloid and antigen-presenting cells, and in particular specialized immune cells in the central nervous system such as microglia. To address this unmet need, we have developed a 29-color panel suitable for spectral flow cytometry, which is able to identify major antigen-presenting cells and myeloid subsets (B cells, DCs, neutrophils, eosinophils, macrophage, monocytes, and microglia) as well as their expression of eight different co-stimulation, adhesion, migratory, and inhibitory proteins. This panel will enable the assessment of these cell types in multiple central nervous system and peripheral tissues in both naïve and disease states.
