Total RNA was extracted using Trizol reagent from the harvested marginal tissues of cerebral infarction, following the manufacturer's procedure. The total RNA quantity and purity were analyzed in Bioanalyzer 2100 and RNA 6000 Nano Lab Chip Kit The sequencing library was constructed with high-quality RNA samples with RIN number > 7.0. The average insert size for the final cDNA libraries was 300 ± 50 bp. Following the vendor's recommended protocol, the 2×150bp paired-end sequencing (PE150) was performed on an Illumina Novaseq™ 6000. The data obtained from sequencing were filtered, subjected to strict quality control, and analyzed using bioinformatics.
