Single-nucleus RNA sequencing of Grn-/- and wild-type mouse thalamus

We performed unbiased nuclei isolation and single-nucleus RNA sequencing of mouse thalamus from progranulin mouse model of fronto-temporal dementia (FTD) (Grn -/-) and wild-type mice. We used 10x Genomics v.2 Single-Cell 3' kit and analyzed 38 samples from four ages: 2 months, 4 months, 7 months, 12 months and 19 months, generating ~200,000 single-nuclei profiles. We identified genes and pathways affected in neuronal and glial cell types in Grn mouse model, which sheds light on cellular and molecular mechanisms of FTD.
