DAP-Seq analysis revealed that AoSte12 was involved in cellular growth and diverse metabolic processes. Overall design: The cDNA sequence encoding protein AoSte12 is constructed in Halo Tag plasmid, the construct vector pAoSte12-halo was identified by sequencing, then the vector and the genome DNA of A. oligospora were send to Genedenovo Biotechnology (Guangzhou, China) for DAP-seq analysis. The AoSte12 protein binds to the target genome fragment, thus the DNA fragment of the genome bound by the target protein can be enriched. Through the combination with high-throughput sequencing technology, DNA products after DAP were sequenced and analyzed, DNA binding sites of target proteins were searched from the whole genome, and high-throughput data results were obtained by efficient sequencing methods. The DNA libraries were sequenced on the Illumina sequencing platform by Genedenovo Biotechnology Co., Ltd (Guangzhou, China).
