Sciatic nerves from P1 and from P60 mice were dissociated, and the resulting cell suspension was sorted with FACS. All cells positive for beta-actin dsred (including Schwann cells also expressing eYFP due to P0Cre-mediated recombination) were collected for RNA sequencing with 10X Genomics. Cells were acutely extracted from P1 and from P60 mouse sciatic nerves derived from beta actin-dsred transgenic animals. The dsred fluorescence allows the selection of dsred positive nerve cells from dsred negative debris in the suspension. Of note, Schwann cells in these experiments additionally express eYFP due to P0cre-mediated recombination. The sorted cells were further processed for library preparation using 10X Genomics. At each age, three independent Chromiun runs were performed, each run loaded with cells derived from independent biological samples. Each biological sample derives from an original cell suspension containing pooled cells extracted from male and female mice in a 1:1 ratio.
