Protein S-glutathionylation, a post-translational modification (PTM), is a dynamic and reversible process forming a disulfide between the glutathione and a cysteine residue of target protein. Through redox proteomic analysis, ARF6 was identified to be glutathionylated on cysteine residue 90 (Cys90). Arf6 is a member of small GTPases ADP-ribosylation factor (ARF) family. C90A mimics the un-glutathionylated form of ARF6. Overexpression of ARF6-C90A increased ferroptosis induced by erastin in H1299 cells. To observe the molecular alterations by ARF6 glutathionylation at Cys90, we performed RNA-seq analysis using ARF6-myc or ARF6-C90A-myc transfected H1299 cells with or without erastin (10 uM) treatment for 12 h.
