Transcription factor Bach1 is a member of the alkaline leucine zipper protein family. Our results therefore highlight a critical role for Bach1 proteins in a regulatory network that integrates essential Wnt/ß-catenin and nodal-Smad signaling to repress mesendodermal differentiation of pluripotent cells Overall design: Taking the advantage of Crispr/Cas9 technique, we establish Bach1 knockout human embryonic stem cell line (Bach1-KO hESCs).The RNA were extracted from WT and Bach1-KO cell lines respectively to detect the markers of the pluripotent gene and three germs layers after spontaneous differentiation . Combined with the results from RNA-seq and CHIP-seq, the role and mechanism of Bach1 in self-renewal and differentiation of human embryonic stem cells were clarified.
