To identify adenosine monophosphate-activated protein kinase (AMPK)-related circular RNA (circRNA) that was differentially expressed in the mouse myocardial ischaemia-reperfusion injury (MIRI) model, and map the AMPK-related circRNA network to provide novel insights for the prevention and treatment of MIRI. Overall design: 32 male C57BL/6 mice were randomly divided into four groups: blank control (Sham) group (n=8), model (MIRI) group (n=8), (MIRI+AICAR) group (n=8), and (MIRI+Compound C) group (n=8). In addition to the Sham group, the other three groups constructed myocardial infarction models by ligating the left anterior descending branch of the mice. After successful modeling, the mice were returned to the thermostat box for 30 min and then the ligature wire was removed to achieve the purpose of myocardial reperfusion, and the mice were continued to be kept for 1 week for the ultrasound examination of the mouse heart, and the mouse heart tissues were obtained for pathological staining, ultramicrotome, serum myocardial markers, and RNA sequencing(RNA-seq), capture differentially expressed circRNA molecules, analyze the main biological processes involved in the differential genes by gene ontology (GO) function, Kyoto Encyclopedia of Genes and Genomes (KEGG) function, qRT-PCR validation of gene RNA-seq sequencing results, and construction of AMPK-related circRNA relationship network.
