We undertook a larger and more ambitious study than the dunes of the Great Western Erg of the Sahara (Algeria) to assess the inter- and intra-site variability of bacterial biodiversity using both methods (suspension-dilution and grain-by-grain), to evaluate the percentage of bacterial culturability using both methods, to build up a collection of species-identified strains and finally to compare this diversity of cultivable bacteria with that obtained by 16S-rDNA metabarcoding.
