This study was undertaken to assess the diversity of endophytic bacterial community prevalent in field shoot-tip tissue of grapevine and the possibility of their in vitro introduction through the explant employing 16S rRNA metagene profiling. Using the shoot-tip derived DNA from grape cvs. Flame Seedless (Sample ID: MG16) and Thompson Seedless (MG17) and the callus tissues generated from the intermodal stem segments of the two cultivars (Sample ID: MG 18 and 19, respectively), 16S rRNA metagene taxonomic profiling was undertaken targeting the V3-V4 hypervariable region employing Illumina MiSeq platform with 2 x 250 bp sequencing chemistry. The samples yielded PE reads in the range of 6,59,153 (MG16) to 11,30,973 (MG17) which after QC and chimera removal were taken through QIIME analysis. This initially showed a significant share of OTUs corresponding to plant chloroplast and mitochondria. Data reanalysis excluding the reads corresponding to plant sequences and the unassigned reads showed high endophytic bacterial community diversity in both the field shoot tips and the aseptically maintained callus tissues.
