In order to discover the cell type in nucleus pulposus and find the cell type specific genetic change during intervertebral disc degeneration, we applied single cell RNA sequencing of nucleus pulposus tissue from degenerated and non-degenerated disc. Overall design: Nucleus pulposus cells are extracted from the tissue immediately after acquire the tissue from surgery. The tissue are then digested using Collagenase for 3 hours. The cell viability are tested and viable sample are send to sequencing.
