A separation–sensing Etalon@lateral flow assay (Etalon@LFA) integrates nafion-functionalized regenerated cellulose (RC) membranes for plasma separation with 3-aminophenylboronic acid (APBA)–functionalized microgel etalon architectures for optical glucose quantification. The device is constructed by spray-coating RC membranes with nafion to form a charge-selective separation layer, followed by the sensing element, a thermoresponsive poly(N-isopropylacrylamide) (pNIPAm) microgel-based etalon structure on RC membranes. Post-processing with APBA renders pNIPAm microgels responsive to glucose through reversible diol–boronate complexation. A single drop of blood-analogue sample, prepared via dispersion of polydimethylsiloxane (PDMS) particles in 4\% sodium dodecyl sulphate (SDS) surfactant solution, is applied to the nafion@RC membrane, enabling plasma separation that subsequently flows into the adjacent etalon@RC sensing region. Glucose binding induces microgel swelling, producing a quantifiable redshift in the corresponding reflectance spectra.
