The single-cell transcriptome profiling of mouse bone marrow LSK cells after resolution of periodontitis

Periodontitis induces myeloid lineage skewing of the bone marrow progenitor cells. In this study, we performed single cell RNA-sequencing in LSK (Lin—Sca-1+cKit+ ) cells to determine whether the periodontitis-induced myeloid lineage bias persists after the resolution of periodontal inflammation. Overall design: C57BL/6 mice were trained by subjecting them to LIP for 21 days followed by ligature removal (to enable inflammation resolution) for 14 days (21dL/14dR, L) or were not ligated during the entire period (NL, N). Hematopoietic progenitors (LSKs; Lin-cKit+Sca1+) in mice bone marrow were sorted by FACS for single-cell transcriptomics analysis.
