We used single cell RNA sequencing to examine the transcriptome of lung-resident memory B cells early after allergen inhalation (day21) and late (day 77) to explore potential mechanisms that control residency in the lungs. Ragweed pollen and OVA were administered to mice intranasally for 5 days in week 1, followed by OVA alone in weeks 2 and 3. Memory B cells were isolated from the lungs by FACS as B220+CD38+GL7-IgD- excluding dead cells (DAPI+) or circulating cells (intravascular label negative) from two mice per time point (day 21 and day 77). 2,500 cells per sample were partitioned on a 10X Chromium Controller and libraries were prepared using 10X Genomics Next GEM Single Cell 3' Reagents Kits v3 (Dual Index). We compared gene expression and memory B cell subsets present on day 21 and day 77 after allergen exposure to identify potential mechanisms controlling residency in the lungs.
