N4-acetylcytidine (ac4C) is a highly conserved RNA modification present in different species. ac4C on mRNA increases the stability of mRNA and the efficiency of protein translation. ac4C has been associated with the development, progression, and prognosis of various human diseases including epithelial ovarian, breast, and bladder cancers. However, the relationship between ac4c and lung cancer has been poorly reported. The mRNA profiling of ac4C in lung adenocarcinoma (LUAD) is unknown. We made the NAT10 expression test and acetylated RNA immunoprecipitation sequencing to identify ac4C peaks on mRNA of human LUAD tissues and adjacent non-tumor tissues, and differences of acetylation and mRNA between the two groups were analyzed. In addition, bioinformatics analysis was conducted to explore the function of ac4C-specific acetylated transcripts. Our study revealed that the degree of ac4C in mRNA in LUAD was significantly higher than in adjacent tissues and was concentrated mainly in the coding sequences with a implications in a wide range of cellular functions.
