Endogenous oxalate synthesis in mouse models of metabolic dysfunction-associated steatotic liver disease- Data Set

The study investigates the effect of diet induced obesity and metabolic dysfunction associated steatotic liver disease induced by a Western diet model on the endogenous synthesis of oxalate in adult male mice. 24-hr Urinary oxalate and glycolate, the activity of the liver enzymes alanine:glyoxylate aminotransferase (AGT), D-glycerate dehydrogenase (DGDH, an accurate index of glyoxylate reductase/hydroxypyruvate reductase or GRHPR), glycolate oxidase (GO), lactate dehydrogenase (LDH), 4-Hydroxy-2-oxoglutarate aldolase (HOGA) were measured. Liver content of oxalate and glyoxylate and kidney oxalate content were measured. Scoring of liver pathology was done using the NAFLD Fibrosis score. C57BL/6J mice fed the high fat diet (60% kcal from fat) reached a mild MASLD stage, C57BL/6N fed the Western Diet (40% kcal from fat, 20% kcal from fructose, 2% cholesterol) reached the moderate MASLD stage. Mice were also administered the endogenous glyoxylate precursor, hydroxyproline intra-peritoneally and urinary excretions monitored after. Urinary oxalate excretion was increased in both groups vs the respective controls, and more so after the hydroxyproline challenge in the Western diet-fed mice group. The activity of liver enzymes involved in glyoxylate metabolism and oxalate synthesis were affected by MASLD indicating greater endogenous oxalate synthesis. The study supports the hypothesis that endogenous oxalate synthesis increases wity the severity of of MALSD in these models. <br>
