In addition to serum immunoglobulins, memory B cell (MBC) generation against SARS-CoV-2 represents another layer of immune protection. We studied longitudinal cohorts of naive individuals and disease-recovered patients to assess the quality of the memory response by analysis of VDJ repertoires, affinity and neutralization against variants of concern (VOCs), using unbiased cultures of single-cell sorted MBCs (Sokal, Immunity 2021, Sokal, Immunity 2022, Sokal JEM 2023, Sokal in preparation). This BioProject groups longitudinal single-cell IGH sequencing sampling data from three cohorts of individuals vaccinated against SARS-CoV-2 using mRNA vaccines (Pfizer-Bio- NTech BNT162b2). - The first cohort consist of mild (n=5) or severe (n=6) COVID-19-recovered patients (first wave, March 2020) subsequently vaccinated with one dose of mRNA vaccine (early 2021). Sampling for this cohort has been done pre-vaccination ("Pre-vac" BioSamples) and 7 days or 2 months post-vaccination ("Post-Vac1" and "Post-Vac2" BioSamples respectively). - The second cohort consist of SARS-CoV-2 naive individuals vaccinated with up to three doses of mRNA vaccine (from early 2021 to early 2022) as part of the French vaccination campaign. Sampling for this cohort was performed at 7 days (plasma cell only; ("Post-Vac1" BioSamples), 2 months ("Post-Vac2" or "D2_M2" BioSamples) and 6 months ("D2_M6" BioSamples) post-2nd mRNA vaccine and one week to two months following a 3rd vaccine dose ("D3_W1-M2" BioSamples). We additionally sampled some of these individuals at 2 months ("PO_M2" BioSamples) and at 6 months ("PO_M6" BioSamples) following an Omicron BA.1 breakthrough infection. - The third cohort consist of patient presenting with various deficiencies in their type-I interferon response (IRF7 LOF (n=2), TLR7 LOF (n=1), IFNAR1 LOF (n=2) or APS-1 with anti-IFN autoantibodies (n=4)) and aged-matched young healthy donors (n=3). Sampling for this cohort was performed between 1 and 6-months post-boost (2nd vaccine dose), as indicated ("D2_M1", "D2_M2", "D2_M3", "D2_M4", "D2_M5" and "D2_M6" BioSamples respectively).
