The trophoblastic disease is a general term for diseases that cause abnormal growth of trophoblasts, the most frequent form of which is called hydatidiform mole (CHM). CHM cell lines (Hmol1-3B and Hmol1-2C) were established in our laboratory (Yamamoto E, et al. Int J Mol Med. 2017 Sep; 40(3): 614â622). In this study, XBP1 was overexpressed, and RNA sequencing was performed to investigate the role of XBP1 in CHM. Overall design: The control and XBP1 lentiviral vectors (pLV[Exp]-Puro-EF1A>EGFP and pLV[Exp]-EGFP:T2A:Puro-EF1A>hXBP1) were purchased from Vector Builder (Chicago, IL). Transduction of the viral vectors was performed according to the protocol of the Lenti-XTM Packaging Single Shots (Takara, Shiga, Japan). Then, the total RNA of cell lines was extracted using the RNeasy Plus Mini Kit (QIAGEN, Hilden, Germany), and RNA sequencing was performed by Novogene, Inc. (Beijing, China).
